Just curious has anyone tried ctb by cutting at the main artery of the neck or is it most likely to fail and if you were to try this with poison would it be more likely to work and kill you faster or is this just not realistic enough

I doubt you'll find anyone here, but people have done so, and with the right determination and power it can work, providing a really sharp knife. BUT only then. So highly risky for most people and likely to fail.

Yes, it's definitely possible. A sharp enough blade and a good amount of force will do it.
A deep enough cut to the carotid artey will result in catastrophic blood loss and rapid death.
