The shortest answer is that it depends on the round being used. Without getting technical, the best bet that's common is going to be 12 gauge, 00 buckshot to the T zone. Chances of survival would be astronomically low with any shot placement to the T zone. Barrel in mouth aiming towards anywhere between 12 o'clock to 3 o'clock (imagine a clock face on a side profile) would do as well, if not better. Closed casket for sure.

if you're looking for an instant death, aim for a shot that will damage the brain stem, it's 100% non-survivable because it regulates almost all daily activities of our bodies like breathing and heartbeat. I'd say aim for the "T-zone" the lower brain rest directly behind it.

Caliber is more important than location. I've heard horror stories of people getting permanent brain damage because they attempted with a .22. Miss the brain stem by a fraction of an inch and you can end up as a vegetable. High caliber is best, or better yet, as others have mentioned, .00 buckshot.

A 3in slug is better than buck at point blank range. But that's purely academic, 2.75 or 3in, slug or 00buck. They are all more than enough, and will all work just fine either in the mouth or heart/sternum.

Regarding caliber, you are 100% correct. One should definitely not attempt with anything smaller than a 9mm, but .40/10mm/.45 would be better for a pistol. .357/.44/.45 for a revolver (no 38 special). Or 7.62/.308 or bigger for a rifle.

These threads are always uncomfortable for me to read. Anyway, whatever you decide. Something like 12 gauge, .00 buckshot. If someone plans to do this, go big either way or stay at home. And I would pick a location where nothing can go wrong.
